553 Multi-omics analysis identifies coordination and hierarchy of transcription factors controlling specific epithelial cell fates in corneal epithelium

چکیده
The homeostasis of the epidermis and corneal epithelium is maintained by proliferation differentiation their corresponding stem cells, keratinocytes in basal layer epidermis, limbal cells limbus cornea. Keratinocytes are morphologically similar but distinct fates to support organ-specific functions. A potential disease mechanism underlying opacity acquiring cell non-transparent emphasizing importance dissect similarity difference between cells. We performed a multi-omics analysis human characterized yet molecular signatures. By gene regulatory network analysis, we identified fate defining transcription factors hierarchy that shared two types those regulate specific epithelial programs. Single-cell RNA-seq cornea confirms factor expression patterns these tissues. Finally, showed genes associated with can cooperatively be targeted cell-specific factors. characterizing signatures, our study uncover circuitry controlling keratinocyte related diseases.

Genome-wide transcription factor (TF) binding profiles differ dramatically between cell types. However, not much is known about the relationship between cell-type-specific binding patterns and gene expression. A recent study demonstrated how the same TFs can have functional roles when binding to largely non-overlapping genomic regions in hematopoietic progenitor and mast cells. Cell-type specif...

Two structurally dissimilar transcriptional regulators, E2A and Pax-5, have been shown to play essential roles in B-cell development; the p50 subunit of NF kappa B, by contrast, is required for normal immune responses.

Objectives Oral squamous cell carcinoma (OSCC) is among the most common cancers worldwide. This study aimed to assess the expression of CK8 and CK17 by OSCC cell lines in Iranian patients. Methods This observational study was conducted on three OSCC cell lines with a minimum of 45 passages. Cells isolated from three patients with OSCC were cultured and passaged. Expression of CK8 and CK17 epi...
متن کاملTranscription Factors Pax6 and AP-2alpha Interact To Coordinate Corneal Epithelial Repair by Controlling Expression of Matrix Metalloproteinase Gelatinase B.
Pax6 is a paired box containing transcription factor that resides at the top of a genetic hierarchy controlling eye development. It continues to be expressed in tissues of the adult eye, but its role in this capacity is unclear. Pax6 is present in the adult corneal epithelium, and we showed that the amount of Pax6 is increased at the migrating front as the epithelium resurfaces the cornea after...
